    How do we solve wicked problems? Effective crowd management

    This paper introduces concepts that may improve our understanding of crowd behaviour and new tools which may help to improve the management of crowds. A multidisciplinary approach taken, drawing on psychology, sociology, mathematics and computer science, amongst other disciplines, and incorporating general management theory, complexity theory and concepts of emergent behaviour and complex adaptive systems. The new tools available for crowd management have been made possible due to advances in digital technology and artificial intelligence

    A roadmap for the future of crowd safety research and practice: Introducing the Swiss Cheese Model of Crowd Safety and the imperative of a Vision Zero target

    Crowds can be subject to intrinsic and extrinsic sources of risk, and previous records have shown that, in the absence of adequate safety measures, these sources of risk can jeopardise human lives. To mitigate these risks, we propose that implementation of multiple layers of safety measures for crowds—what we label The Swiss Cheese Model of Crowd Safety—should become the norm for crowd safety practice. Such system incorporates a multitude of safety protection layers including regulations and policymaking, planning and risk assessment, operational control, community preparedness, and incident response. The underlying premise of such model is that when one (or multiple) layer(s) of safety protection fail(s), the other layer(s) can still prevent an accident. In practice, such model requires a more effective implementation of technology, which can enable provision of real-time data, improved communication and coordination, and efficient incident response. Moreover, implementation of this model necessitates more attention to the overlooked role of public education, awareness raising, and promoting crowd safety culture at broad community levels, as one of last lines of defence against catastrophic outcomes for crowds. Widespread safety culture and awareness has the potential to empower individuals with the knowledge and skills that can prevent such outcomes or mitigate their impacts, when all other (exogenous) layers of protection (such as planning and operational control) fail. This requires safety campaigns and development of widespread educational programs. We conclude that, there is no panacea solution to the crowd safety problem, but a holistic multi-layered safety system that utilises active participation of all potential stakeholders can significantly reduce the likelihood of disastrous accidents. At a global level, we need to target a Vision Zero of Crowd Safety, i.e., set a global initiative of bringing deaths and severe injuries in crowded spaces to zero by a set year

    Current strategies for quantification of estrogens in clinical research

    Estrogens and their bioactive metabolites play key roles in regulating diverse processes in health and disease. In particular, estrogens and estrogenic metabolites have shown both protective and non-protective effects on disease pathobiology, implicating the importance of this steroid pathway in disease diagnostics and monitoring. All estrogens circulate in a wide range of concentrations, which in some patient cohorts can be extremely low. However, elevated levels of estradiol are reported in disease. For example, in pulmonary arterial hypertension (PAH) elevated levels have been reported in men and postmenopausal women. Conventional immunoassay techniques have come under scrutiny, with their selectivity, accuracy and precision coming into question. Analytical methodologies such as gas and liquid chromatography coupled to single and tandem mass spectrometric approaches (GC–MS, GC–MS/MS, LC–MS and LC–MS/MS) have been developed to quantify endogenous estrogens and in some cases their bioactive metabolites in biological fluids such as urine, serum, plasma and saliva. Liquid-liquid or solid-phase extraction approaches are favoured with derivatization remaining a necessity for detection in lower volumes of sample. The limits of quantitation of individual assays vary but are commonly in the range of 0.5–5 pg/mL for estrone and estradiol, with limits for their bioactive metabolites being higher. This review provides an overview of current approaches for measurement of unconjugated estrogens in biological matrices by MS, highlighting the advances in this field and the challenges remaining for routine use in the clinical and research environment

    Background: Cellulose-based materials have been used for centuries to manufacture different goods derived from forestry and agricultural sources. In the growing field of nanocellulose applications, its uniquely engineered properties are instrumental for inventive products coming to competitive markets. Due to their high aspect ratio and stiffness, it is speculated that cellulose nanocrystals (CNC) may cause similar pulmonary toxicity as carbon nanotubes and asbestos, thus posing a potential negative impact on public health and the environment. Methods: The present study was undertaken to investigate the pulmonary outcomes induced by repeated exposure to respirable CNC. C57BL/6 female and male mice were exposed by pharyngeal aspiration to CNC (40 ÎŒg/mouse) 2 times a week for 3 weeks. Several biochemical endpoints and pathophysiological outcomes along with gene expression changes were evaluated and compared in the lungs of male and female mice. Results: Exposure to respirable CNC caused pulmonary inflammation and damage, induced oxidative stress, elevated TGF-ÎČ and collagen levels in lung, and impaired pulmonary functions. Notably, these effects were markedly more pronounced in females compared to male mice. Moreover, sex differences in responses to pulmonary exposure to CNC were also detected at the level of global mRNA expression as well as in inflammatory cytokine/chemokine activity. Conclusions: Overall, our results indicate that there are considerable differences in responses to respirable CNC based on gender with a higher pulmonary toxicity observed in female mice
